UTICAJ GENISTEINA I ESTRADIOLA NA JAJNIKE NEZRELIH PACOVA.
STEREOLOŠKA ANALIZA
The effects of genistein and estradiol on the ovaries of immature rats. Stereological study.

Rezime
Genistein je supstanca prirodnog porekla, sa estrogenskom aktivnošdu, prisutan je u supstancama
prirodnog porekla koje se koriste u ishrani. Cilj ovog rada je da ispita uticaj genisteina na stereološke
karakteristike jajnika polno nezrelih pacova. Efekat genisteina uporedjen je sa efektom sintetičkog
estrogena 17α-etinil-estradiola. Šest 18 dana starih ženki pacova davano je 35 mg/kg genisteina (G),
rastvorenog u DMSO, jednom dnevno, tokom tri dana. Drugoj grupi ženki subkutano je aplikovan 1 µg/kg
17α-etinil-estradiol (E), rastvorenog u maslinovom ulju u istoj zapremini. Svaka eksperimentalna grupa
imala je svoju kontrolnu grupu, koja je primala istu količinu DMSO (CG) ili maslinovog ulja (CE).
Genisteinski i estradiolski tretman nisu uticali na promenu telesne mase, apsolutnih i relativnih masa
jajnika. Volumen folikularnog parenhima (V fp), volumen ovarijalne strome (V os) and i ukupni volumen
jajnika (V ov), su određevani pomodu Kavalirijevog principa. V fp je ostao ne promenjen nakon genisteinskog
tretmana, dok se V os povedao za 53.40% (p<0.05). Nakon izlaganja estradiolu, V fp se smanjio za 34.17%
(p<0.05), dok je V os povedan 34.17% (p<0.05). Na osnovu naših rezultata možemo zaključiti da je
genistein, kao i estradiol, poremetio odnos folikularni parenhim-ovarijalna stroma, izazivajudi povedanje
volumena V os. Nasuprot estradiolu, genistein je doveo do povedanja V os, ali nije uticao na V fp.

Summary
Genistein is plant-derived compound, with estrogenic activity, found in natural dietary sources. The aim
of the present study was to investigate the effect of genistein exposure on the stereological
characteristics of ovaries of immature rats. The effects of genistein were compared with the influence of
synthetic estrogen, 17α-ethinyl-estradiol. Six 18-day-old females subcutaneously received 35 mg/kg of
genistein (G), dissolved in dimethylsulfoxide (DMSO), daily for 3 days. Second group of females
subcutaneously received 1 µg/kg of 17α-etinil-estradiol (E) dissolved in olive oil by the same schedule.
Each experimental group had a corresponding control group, receiving the same volume of DMSO (CG),
or olive oil only (CE). Genistein and estradiol treatment did not change body weight, absolute and
relative ovary weight. The volume of follicular parenchyma (V fp), volume of ovarian stroma (V os) and total
ovary volume (V ov), were estimated using Cavalieri’s principle. The V fp was remained unchanged after
genistein treatment, while V os increased by 53.40% (p<0.05). After exposure to estradiol, V fp decreased by
34.17% (p<0.05), while V os increased 34.17% (p<0.05). On the basis our result we can conclude that
genistein, as estradiol, disturbs the follicular parenchyma-ovarian stroma ratio, inducing an increase of
V os. In contrast to estradiol, genistein increased V os, and did not affect V fp.
