POVECANJE KONCENTRACIJE HAPTOGLOBINA I ALFA2-MAKROGLOBULINA U

                            SERUMU PACOVA TOKOM AKUTNO FAZNOG ODGOVORA


                      THE INCREASE OF HAPTOGLOBIN AND ALPHA2-MACROGLOBULIN
                  CONCENTRATIONS IN RAT SERUM DURING THE ACUTE PHASE RESPONSE

                                                           REZIME

               Različite vrste povreda pokredu univerzalan i evolutivno očuvan mehanizam odbrane organizma poznat
               kao akutno fazni odgovor (AFO). U ovoj sistemskoj reakciji organizma važnu ulogu ima jetra, u kojoj se
               sintetše grupa proteina, poznatih kao akutno fazni proteini (AFP). Dva najznačajnija AFP kod pacova su
               alfa 2-makroglobulin (MG) i haptoglobin (Hp). Cilj ovog rada je bio da se metodom imunoelektroforeze
               odredi  relativna  količina  MG  i  Hp  u  serumu  pacova  nakon  inducije  AFO  terpentinom,  kao
               eksperimentalnom modelu AFO. Pradenjem kinetike (6, 12 i 24 sata nakon indukcije AFO), uočava se da
               se  relativna  količina  oba  izučavana  AFP  povedava  sa  vremenom  nakon  indukovane  povrede,  da  bi
               maksimalan nivo bio zabeležen u 24. satu. To je u skladu sa literaturom, jer od 24 do 48 sata nakon AFO
               dostiže plato AFP u cirkulaciji, nakon čega opada. U tom periodu, MG, kao protein koji inhibira proteaze
               oslobođene  iz  delija  na  mestu  povrede,  štiti  tkiva  od  dalje  destrukcije,  a  Hp  sakuplja  oslobođeni
               hemoglobin iz liziranih eritrocita i sprečava dalji gubitak gvožđa iz organizma.

                                                          ABSRACT

               Different  types  of  trauma  induce  universal  and  evolutionary  conserved  protective  mechanism  in
               organism known as acute phase response (APR). During this systematic reaction liver has important role
               producing  set  of  liver-derived  proteins,  termed  acute  phase  proteins  (APP).  Two  most  important
               members of APP in rat are alpha2-macroglobulin (MG) and haptoglobin (Hp). The aim of this work was to
               determinate  the  relative  MG  and  Hp  concentrations  in  rat  serum  after  turpentine  APR  induction  by
               immnuoelectrophoresis. The results revealed the time-course increase of MG and Hp concentrations in
               rat serum after APR induction. The highest level was observed 24 hours after turpentine treatment. This
               result is in correlation with literature data showing the highest level of APP in circulation 24-48 hours
               after the induction of APR, when the APP concentrations decrease. In this period, MG, as inhibitor of
               proteases  released  from  the  injured  cells,  protect  tissue  from  further  destruction.  The  role  of  Hp  is
               binding of hemoglobin after haemolysis in order to prevent the loss of iron from organism.
