                    TUMOR SUPRESSOR P53 IN ACUTE PHASE RESPONSE AND ACUTE RADIATION

                                                          REZIME


               Akutno fazni odgovor (AFO) predstavlja ranu i nespecifičnu reakciju organizma na različita oštedenja tkiva i infekcije
               koja ima za cilj ponovno uspostavljanje narušene homeostaze.  Izlaganje celog organizma jonizujudem zračenju u
               dozama  višim  od  1  Gy  dovodi  do  oboljenja  koje  se  ispoljava  oštedenjem  različitih  tkiva  i  organa  poznatom  pod
               nazivom akutna radijaciona bolest (ARB). Protein p53 je transkripcioni faktor koji učestvuje u regulaciji različitih

               gena čiji su produkti direktni i/ili indirektni medijatori delijskih mehanizama, koji se aktiviraju u različitim vrstama
               stresa. Cilj ovog rada je bio da se metodom imuno-blot analize odredi prisustvo relativne količine proteina p53 u
               jedarnim proteinima jetre pacova tokom AFO i ARB. Rezulati su pokazali da se količine p53 u jetri pacova nakon AFO

               slabo indukuje u poređenju sa kontrolom, dok je povedanje ovog proteina tokom ARB značajno uvedano. Razlika u
               količini p53 u AFO i ARB je upravo razlika u prirodi oštedenja koja se javljaju u ova dva model sistema povrede
               organizma.  Oštedenje  DNK  je  jedan  od  najintenzivnijih  stimulatora  p53,  a  ova  oštedenja  su  značajna  posledica
               izlaganja organizma zračenju, što se reflaktuje i na relativnu količinu proteina p53 u jetri tokom AFO i ARB.

                                                        ABSTRAKT



               The acute phase  response (AFR) represent an early and nonspecific response of organisam on different kind of
               tissue injures and infections with the aim to restore disturbed homeostasis. Exposure of organism to irradiation
               higher than 1 Gy leads to differnt degrees of tissue and organ injures known as acute radiation desease (ARD).
               Protein p53 is transcription factor that regulates different genes that are activated in different types of stress. The

               aim of this work was to determina te the relative amount of p53 in rat liver nuclear proteins during APR and ARD by
               immuno-blot analysis. The results revealed that the induction of p53 during APR is slightly higher compared to the
               control  level,  while  the  ARD  induced  significant  increase  of  p53.  APR  and  ARD  represents  two  different  model

               systems of trauma and different expression of p53 is the consecuense of differences in the degree of injury during
               APR and ARD. DNA damage is one of the most important inducer of p53 expression and DNA damage is very high
               after irradiation exposure that reflects on the level of p53 during APR and ARD.
