IZOLOVANJE DIARILHEPTANOIDA IZ BILJNE VRSTE

                                                          REZIME


                      U ovom radu je prikazano izolovanje i odreĎivanje diarilheptanoida oregonina i metilhirsutanonola iz kore
               drveta biljne vrste  Alnus glutinosa (jova). Poznato je da kora drveta jove sadrži diarilheptanoide, jedinjenja koja
               pokazuju  antibakterijsko,  anti-inflamatorno,  antivirusno  dejstvo  na  virus  gripa  i  antioksidativno  dejstvo.  Ova

               jedinjenja su našla značajnu primenu i u hemoterapiji. Ekstrakcija kore drveta jove je vršena tri puta, pomoću sistema
               rastvarača metanol/hloroform (50:50). Dobijeni ekstrakt je razdvajan gradijentnim programom na koloni silika gela
               sistemom  rastvarača  metilen-hlorid/metanol.  Tok  razdvajanja  je  praćen  tankoslojnom  hromatografijom  na
               aluminijumskim  pločicama  obloženim  silika  gelom  dimenzija  20×20  cm.  Na  osnovu  dobijenih  Rf  vrendosti    su
               formirane zbirne frakcije od kojih su analizirane AG 14-30 i AG 140-152. Zbirna frakcija AG 14-30 je razdvajana

               pomoću semipreparativnog HPLC-a,  po programu koji je dao 8 frakcija i posle snimljenih NMR spektara utvrĎeno
               je da je prva frakcija AG 14-30/1 čisto jedinjenje-metilhirsutanonol. Zbirna frakcija AG 140-152 je prečišćavana
               preparativnom  tankolsojnom  hromatografijom  na  razvučenim  pločama  silika  gela.  Snimljeni  su  NMR  spektri
               prečišćene frakcije i njihovom analizom spektara identifikovan je diarilheptanoid oregonin.

                                                         SUMMARY

                      This   research   was   focused   on isolating and   determination   of   diarylheptanoids,   oregonin
               and metilhirsutanonol from   the   bark   of Alnus glutinosa (alder)   plant   species. It   is   known   that
               alder bark contains diarylheptanoids                        that show antibacterial, anti-inflammatory,

               antiviral   effect on influenza virus, and   antioxidant   activity. These   compounds   have found
               considerable application  in chemotherapy. Extraction  of the  bark  of alder  was  performed three  times, using
               a solvent system methanol/chloroform (50:50).           The                  obtained extract was
               separated   by gradient program   on   the   silica   gel column in solvent system   methylene   chloride-
               methanol. Flow separation was  monitored by  thin  layer  chromatography  on aluminum  plates  coated with  silica
               gel dimensions of 20 ×20 cm. On the basis of  Rf  values are formed multifractions of which are analysed AG 14-30

               and AG 140-152. Multifraction AG 14-30 was separated by semipreparative HPLC, according to the program which
               gave 8 fractions and after recorded NMR spectra  it was clear that first fraction, AG 14-30/1 was pure compound–
               methylhirsutanonol.   Multifraction  AG 140-152 was separated  by  preparative thin  layer chromatography  on silica
               gel plates.  NMR spectra of  rafined  fraction  were recorded  and  after  their  analysis  diarylheptanoid  oregonin  was
               identified.
